Abstract

¿Ü»ó¼º ¼Õ»ó ÈÄ Ä¡¾Æ Å»±¸ ½Ã Ä¡¾Æ¸¦ ÀúÀåÇÏ´Â ÀúÀå¿ë¾×ÀÇ Á¾·ù¿Í ¿Âµµ°¡ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²¿¡ ¾î¶°ÇÑ ¿µÇâÀ» ¹ÌÄ¡´ÂÁö ¿¬±¸Çϱâ À§ÇÏ¿© Ä¡ÁÖÀδ뼼Æ÷¸¦ 10% fetus bovine serum(FBD) ÇÔÀ¯ ¥á-minimal essential medium(¥á-MEM)¿¡¼­ 37¡É 5% CO_(2) °ø±â È¥ÇÕ ¹è¾ç±â¿¡¼­ ¹è¾çÇÏ°í 4.25, 37¡ÉÀÇ Hank¡¯s balanced sa1t solution(HBSS)°ú ¥á-MEM, ¿ìÀ¯(Sȸ»ç, Pȸ»ç), tap water¿¡ ÀúÀåÇÏ°í 60ºÐÀÌ Áö³­ ÈÄ °¢ ±º¿¡ ´ëÇØ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²À» ÃøÁ¤Çϱâ À§ÇÏ¿© MTT assay¸¦ ½ÃÇàÇÏ¿© ´ÙÀ½°ú °°Àº °á°ú¸¦ ¾ò¾ú´Ù.

1. 4¡É ÀúÀå¿ë¾×ÀÇ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²Àº ¥á-MEM°ú Pȸ»ç¿ìÀ¯¿¡¼­ °¡Àå ³ô¾Ò°í HBSS, Sȸ»ç¿ìÀ¯, tap water ¼øÀ¸·Î ³·¾Ò´Ù.

2. 25¡É ÀúÀå¿ë¾×ÀÇ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²Àº ¥á-MEM¿¡¼­ °¡Àå ³ô¾Ò°í Pȸ»ç¿ìÀ¯, HBSS, Sȸ»ç¿ìÀ¯, tap water ¼øÀ¸·Î ³·¾Ò´Ù.

3. 37¡É ÀúÀå¿ë¾×ÀÇ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²Àº ¥á-MEM°ú Pȸ»ç¿ìÀ¯, HDSS, Sȸ»ç¿ìÀ¯¿¡¼­ ³ô¾Ò°í tap water¿¡¼­ °¡Àå ³·¾Ò´Ù.

4. ¥á-MEMÀÇ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²Àº 4¡É¿¡¼­ °¡Àå ³ô¾Ò°í 25¡É¿Í 37¡É ¼øÀ¸·Î ³·¾ÒÀ¸¸ç HBSS¿¡¼­´Â 4¡É¿¡¼­ ³ô¾Ò°í 25¡É¿Í 37¡É¿¡¼­ ³·¾Ò´Ù.

ÀÌ»óÀÇ °á°ú¸¦ Á¾ÇÕÇغ¼ ¶§ ¿Ü»óÀ¸·Î ÀÎÇØ Ä¡¾Æ°¡ Å»±¸µÇ¾úÀ» ¶§ Å»±¸µÈ Ä¡¾ÆÀÇ ÀúÀå¿ë¾×À¸·Î HBSS ¿ë¾×ÀÌ ÃßõµÇ°í ÀÖÀ¸³ª ÀÌ ¿¬±¸¿¡¼­ 4¡É¿Í 25¡É ¿ìÀ¯¿¡¼­ Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÁ¸À²ÀÌ ³ô¾ÒÀ¸¹Ç·Î »ç°í ÇöÀå¿¡¼­ ½±°Ô ±¸ÇÒ ¼ö ÀÖ°í Ä¡ÁÖÀδ뼼Æ÷ÀÇ »ýÈ°·Â º¸Á¸¿¡µµ À¯¸®ÇÑ ³·Àº ¿ÂµµÀÇ ¿ìÀ¯¿¡ Å»±¸µÈ Ä¡¾Æ¸¦ º¸Á¸ÇÏ´Â ¹æ¹ýµµ ÁÁÀ» °ÍÀ¸·Î »ý°¢µÈ´Ù.

To compare the survival rate of periodontal ligament cells preserved in storage media with good availability at the time of an avulsion injury, periodontal ligament cells were incubated in ¥á-MEM culture medium containing 10% FBS in condition of 37¡É, 5% CO_(2). These cells were then cultured in HBSS, ¥á-MEM, milk(S. co., P. co.) and tap water at the temperature of 4, 25, 37¡É each in 60 min. The groups were measured by MTT assay.

The results were as follows :

1. Among the storage media at 4¡É, ¥á-MEM and P-milk had the highest preserving ability of periodontal ligament cells, while that of HBSS, S-milk and tap was low in order.

2. Among the storage media at 25¡É, ¥á-MEM had the highest preserving ability of periodontal ligament cells, while that of P-milk, HBSS, S-milk, tap water was low in order.

3. Among the storage media at 37¡É, the preserving ability of periodontal ligament cells was very high in ¥á-MEM, P-milk, HBSS and S-milk. it¡¯ s lowest in tap water.

4. The preserving ability of periodontal ligament cells in ¥á-MEM was high at 4¡É and it¡¯s low in order of 25¡É, 37¡É, but in HBSS was high at 4¡É and it¡¯s low at 25¡É, 37¡É.

5. The preserving ability of periodontal ligament cells in S-milk and P-milk was high at 4¡É, 25¡É and it¡¯s low at 37¡É.

In conclusion, HBSS is the storage medium of choice in an avulsion, but in this study it is preferable to choose milk at 4¡É for tooth since it is easy to get and affect cell viability
